Effects of combined antibiotics against gentamicin-resistant Staphylococcus aureus

Abstract

|
Nine strains of Staphylococcus aureus resistant to gentamicin (GM) were tested for the combined action of GM with methicillin (MT), tetra cycline (TC), streptomycin (SM), cephaloridine (CP), sulbenicillin (SB), and carbenicillin (CB).
Population analysis in the presence of increasing concentrations of GM by tube dilution method showed that the number of colony forming unit (CFU) per milliliter after an 18 hr incubation was 104 to 107 at each minimal inhibitory concentration (M IC), and mostly became fewer than 10at the concentrations of twofolds or four-folds of MIC of each drug The combined action of antibiotics could be expressed by the value of ? (MIC of GM alone/MIC of GM with 1/4 of MIC of other antibiotic). The values of mean ? MT, ?TC, ? SM, ?CP, ? SB, and ?CB, were 18.7, 2.6, 2.7, 21.3, 14.0, and 14.0, respectively. Thus, the combined action of GM with penicillins or CP was high, particularly in GM?CP combination, while low with TC or SM.
When determined by the agar dilution method, synergic effect of GM?MT was noted in 4 strains, while GM?TC did not show any such effect on all of the strains tested.
Time-killing curves were plotted on the strains which were greater than 4 in value of ?, regardless of antibiotic combination. The number of CFU on the media containing combined drugs with 1/4 of the MIC of each drug resulted in less than one-hundredth than each drug alone at 3 hr incubation. At 6 and 12 hr incubations, the number of CFU on the media containing penicillins or CP decreased more markedly, while the numbers increased on the media of GM.TC and GM.SM. At 18 hr incubation, there were from 3.4X 104 to 107 CFU/nt in GM?TC and GM?SM, while fewer than 104 CFU/ml in the combination of GM and penicillins or CP.
